"What is the most rewarding part of this position?"
"What options are available for professional development?"
"What do you think may be a challenge in transitioning to this position" NOTE: Always respect their opinion, but subtlely rebuttal it and find a way that you can make that transition.

I have been on both sides of the fence in job interviews, and I like to hear and ask questions about how the applicant can improve the situation. So for example, "What is/are the biggest challenge(s) facing this position/department?" Most companies/managers are mostly concerned about how you will make/save them money and time. If you can alleviate the problems they are currently facing you make them happy. Happy people like to give you more promotions and raises.

It's important to understand the hiring process prior to asking this question. For instance, many jobs will require that you go through a series of interviews, recruitment events, tests, etc. prior to making you an offer. I am aware of at least one person that went through six separate interview stages for one position. Obviously, asking a question about when you would start in the position would be inappropriate during the first interview stage for this case. However, if you ask when they want you to start at the right time, it could go over well.
