Gamesprays Zombie Edition!
The Zombie edition of Gamesprays.com is finally live, like 15 minutes ago. Was a blast to design. I'll be making sprays in spare time to get it rolling like I did for the TF2 edition. The current sprays are submitted by members of the TF2 edition. Should become a decent spot to find and share L4D sprays.
Gamesprays is a side project that I work on in my spare time. If you find any bugs, have any suggestions or feedback, feel free to pm me or post here.
Ok, I need a break. My BRAAAAAINS are tired.

I don't have a tutorial on making sprays specifically for L4D yet but I'm sure my tutorial on making them for TF2 will work for L4D since they use the same system and final .vtf spray format. Anyway, here is the TF2 spray tutorial.

How to make a spray:
1. Open paint, photoshop, or whatever.
2. Draw and edit to your hearts content.
3. Save as jpg.
4. Run the game and go into settings/options.
5. Find the place in there that lets you change your spray.
6. Click Browse and browse for your spray.
*Or you can just start from 4 and use any picture on your computer.

Get the VTF Plugin
Good advice. Also I highly recommend downloading the VTF plugin for Photoshop or Paint Shop Pro. Besides making it really easy to save in .vtf format (no need to open another program ) It will warn you if you try to save with invalid dimensions AND you can select a preset of "Spray with Alpha" making it a no brainer to get the right settings.
I've made several L4D sprays with it. By far, my favorite is: Shaun of the Dead Against a Window.
