I'm having the same problem, but it only started yesterday.
A few days ago, I updated my video drivers and that's about all I've changed. I may try to reinstall the game because it worked just fine since launch till yesterday.
System specs if that makes a difference:

Just wanted to add an update:
I just tried playing Counter-Strike: Source and Team Fortress 2 with no problems so I don't think it is the Source Engine or Steam.
It usually happens to me in L4D only and it freezes for about 10 seconds every 5-10 minutes.
I'll reinstall later when I have time and post any updates as needed.
Another edit:
After some reading on the steam forums I found this:

Seems like a good thing to follow...

This game is really startin to suck
i have uninstalled and installed this game 5 time. i have downloaded new drivers for my video card, my audio card and my motherboard. i have set the audio and video settings from high to low. i have a 3000 dollar gaming machine from Digital Storm Corp. replaced the geforce 8800gts with a geforce 260gtx 896mb ddr3. and it still freezes for a few seconds off and on. if they dont release a patch or something soon for this problem, im goin return this game.

Click on Advanced Settings or Advanced Options to display more complex graphical settings. In there you should see all available graphics settings to tweak.
